    Antioxidant

    Phloretin has strong antioxidant properties and scavenges free radicals in the skin. Rhizopusin, the glucoside of rhizopus, has also been shown to have antioxidant activity, with 3-hydroxyrhizopus being able to scavenge peroxides, free radicals, and lipid peroxidation. Rhizopusin inhibits the release of nitric oxide from stimulated macrophages and reduces macrophage phagocytosis, with potential anti-inflammatory effects.

     

    Anti-Inflammatory

    Phloretin and epicatechin are the strongest inhibitors of cyclooxygenase 1, and rhizopus is a known competitive inhibitor of D-glucose co-transporter proteins, giving rhizopus a potential anti-inflammatory effect.

     

    Sun Protection

    Phloretin has a certain UV-absorbing capacity. Phloretin absorbs UV rays directly but also enhances the expression of nucleotide excision repair genes. It slows down UVB-induced pyrimidine dimer formation, glutathione degradation, and cell death, reducing UV damage to keratinocytes.

     

    UV irradiation can cause cells to produce reactive oxygen species and free radicals, both of which are extremely unstable and prone to a series of oxidative chain reactions that ultimately damage cells. Adding rhizomes to cosmetic base formulations can increase the SPF and SPA values of cosmetics. In addition, a mixture of rhizomes, VC, and ferulic acid can protect human skin from UV rays and provide photoprotection for human skin.

     

    Whitening and Anti-Aging

    Phloretin inhibits melanocyte activity and has potential whitening effects. It has been found that 3-hydroxyrhizoderma shows potent antioxidant and tyrosinase-reducing effects in human epidermal melanocytes. The powerful antioxidant activity gives rhizopodophyllin an anti-aging effect. Phloretin analogs also have inhibitory activity on xanthine oxidase and elastase, which can inhibit the synthesis of MMP-1 in human fibroblasts and reduce the decomposition of collagen and other components, thus inhibiting skin aging.

     

    Bacteriostatic

    Phloretin is a flavonoid compound with antibacterial activity. It has been found that phloretin has a strong inhibitory effect on Gram-positive bacteria, especially on Staphylococcus aureus, Listeria monocytogenes and Salmonella typhimurium. In addition to its bacteriostatic activity against pathogenic bacteria, rhizopin is also useful against common skin bacteria.

     

    The MIC of phloretin on both Propionibacterium acnes and Propionibacterium granulosum was 0.5mg/L. Meanwhile, the results of the clinical trial showed that there was a significant reduction of whiteheads, blackheads, pimples, and sebum secretion in the subjects after usingphloretin for 4 weeks. The inhibitory effect of phloretin on bacteria is on the one hand to act on the bacterial internal enzyme protein system, affecting the bacterial energy metabolism and bacterial growth of oxidative damage repair; on the other hand, it interferes with the synthesis of intracellular proteins, so that the metabolism of the cell is disturbed, which ultimately leads to the death of the bacteria.

     

    Transdermal Permeation

    Phloretin has a very safe and effective transdermal permeability effect, which enhances the transdermal permeability of drugs. Multiple studies have confirmed that Phloretin and 6-ketocholanol interact with the lipid layer and alter the structure of the cell membrane, resulting in greater fluidity. As a transdermal permeation enhancer, rhizopin can facilitate the absorption and utilization of other potent ingredients in the formulation, making them more effective.
