    Inhibiting Tumour Growth

    A study reported in Nature 2018 found that D mannose powder can inhibit tumour growth by suppressing glucose metabolism by screening the effects of different hexoses on the growth of tumour cell lines. Lui Lei's team sought to explore whether various hexoses affect tumour immunity. Through screening, the researchers found that high concentrations of D-mannose were able to degrade PD-L1 protein in TNBC cells. In-depth mechanistic studies revealed that D-mannose treatment could promote the activation of AMPK, which promoted the phosphorylation of the S195 site of PD-L1 protein. The phosphorylation of this site can weaken the glycosylation of PD-L1 protein, which leads to the degradation of PD-L1 protein through proteasome.

     

    Further studies revealed that D-mannose-mediated degradation of PD-L1 also inhibited the DNA damage repair process in tumour cells, resulting in a facilitating effect on radiotherapy for TNBC. In a mouse model of TNBC, the combination of oral D mannose and PD-1 antibody treatment significantly prolonged the survival of hormonal mice and increased CD8+ T cell infiltration and granzyme B expression in the tumour. Similarly, the combination of oral D-mannose and ionising radiation significantly inhibited tumour growth in vivo and significantly enhanced the efficacy of radiotherapy in TNBC mice.

     

    In conclusion, this study revealed a new function of D-mannose - degradation of PD-L1 protein and reported for the first time the facilitating effect of D-mannose on immunotherapy and radiotherapy of TNBC, which provides a new strategy for the clinical treatment of TNBC.
